Easy to find, but fun :

- Commemorative challenge : you can only use cards with flashback and creatures with persist (and lands).

- loud mana challenge : you can only use cards with at less 2 mana of the same color in the CC (and lands).

An hard one :

Give me your soul challenge : each turn, AI can play a random card of your deck without pay the mana cost.
(should be a difficult challenge, I wouldn't play it !...).

End game challenge : in this challenge, AI and players have a lot of permanents into play. When game start, it look like a 5 or 6 or more turns beginned game with complex interactions.
Difficulty is increased by fact that player can't play colorless spell (artifacts or eldrazi), and each game player start with differents lands into play that give differents colors, or sometimes permanents that are more an handicap than a advantage. So the player deck must be very adaptative.
There is a puzzling part in this challenge.

There can be only one 2 : in this challenge, player can only put card with a casting cost of 1 in his deck (and lands).

Big story challenge : Player can only use cards with flavor text > 3 lines. (And basic lands and duals lands)

To be tree : cards of player must have exactly 3 colored mana (and any coloreless mana). +lands.

Masochist challenge : if during his turn, player don't lose life, he loose the game.

(so first turn is very important, player must play something like mogg fanatic, sac it and hurt himself, or play a spell making him loose life like gitaxian probe).

Near the same challenge :
if during the turn of the player, AI don't gain life, player loose the game.

The Legend : player can only use Legendary permanents and cards restricted to one. +basic and dual lands.

For more difficulty, cards restricted to one can be removed, so player can only play legends.

Super drawing man : if during his turn, player don't draw at least 2 cards (draw step + 1 more card), he loose the game.

More difficult and near the same :
If player don't play 2 or more spell during his turn, he loose the game.

Exiled : each turn, any number of random card of player library are exiled.
Number of cards exiled can increase each turn.
Additionnal rule : player deck must be exactly 60 cards.

Artist challenge : an idea I have posted before, but not well.
Except basic and dual lands, total number of artists of player cards can't be more than 4.

Maybe difficulty can be balanced, restrict number of artist to 3, or expand it to 5.

Big feeder : AI can't lose the game. But if AI reach 50 life or more, AI lose the game.

Big teatcher : AI can't lose the game. But if AI reach 15 or more cards in hand, AI lose the game.
(should be easy with jace or properity).

Extra capacity : player can only play cards with a minimum of 2 colored mana symbols in card text. (+ lands)

Bad confidant : each time player draw a card, he loose life = CC of this card.

There can only be one 3 : at end of his turn, player discard all cards except one.

Tap me : player can only play cards with the "tap" symbol in the text.

Pacific dude : player can only play creatures cards with power 0 (+basic and dual lands).

Coallition deck : deck must contains at least 15 cards that are multicolored and have 5 colors. Player can only play basic lands.

No draw No go : except the card he draw during his draw step, player can't draw cards. Player can't play any spell or capacity during AI turn.

Mana source : player can only play cards that give mana or have a mana source capacity (like llanowar elfe).

Unless otherwise noted, all these deck construction challenges allow alpha/beta lands as usual.

Get Equipped!: Your deck must consist entirely of equipment cards. You start three 0/1 kobolds in play (or something similar.) Alternatively, it could allow six cards that break the rule, but something about starting with a few 0/1 creatures seems more interesting.

Auramancy: Every card in your deck must be an aura. You may have six cards that break this rule.

Power of One: Every card in your deck must have a CMC of exactly one.

Mirrored Colors: Your deck cannot have any mana sources (even the beta lands are banned), nor any artifacts or colorless cards -- with the exceptions of Exotic Orchard, Fellwar Stone, Quirion Explorer, Sylvok Explorer, and Benthic Explorers (that is, mana sources that can only produce colors of mana your opponent can produce.) You may have any number of copies of Exotic Orchard in your deck.

(Alternative to the above:)
Mad Lands: At the start of each duel, the game chooses from 1-5 colors at random, then replaces all lands in your deck with random basic lands of those colors. You may not use any artifacts or colorless cards.

Unrestricted: You may ignore all limits on deck construction, save that you must have at least 60 cards. Your opponent starts with 60 life and a Chromatic Lantern in play, and draws an extra card each turn. (Probably your opponent needs more of an advantage for this to be fair.)

Balanced Out: Your deck must have an equal number of mono-color cards of each color. No gold cards are allowed, and aside from alpha/beta lands, no colorless cards, either.

Paralyzed: Skip your untap phase. (This might be too harsh to be winnable; so perhaps allow them to untap one basic land per turn, or even just allow their basic lands to untap normally? On the other hand, it's fun to think of ways to beat it when you get no untap phase at all and no compensation.)

Fading...: Every card in your deck must have Fading, Cumulative Upkeep, Phasing, or Echo.

Junkyard: Excluding beta lands, your entire starting hand and all cards you draw from your deck are immediately discarded.

Repetition: Each round, half the cards in your deck are replaced by copies of one particular card. (Eg. half your deck is replaced by Goblin Welders one round; Clones another, etc etc etc.)

It would be cool to get an EDH gauntlet, where the opponent gets moxen or something. Basically anything edh would be fun.
